Maria Shriver and Caroline Kennedy are reported to have lobbied hard for the History Channel to pull the plug on the series.
The eight part film, which stars Katie Holmes as First Lady Jackie Kennedy and Greg Kinnear as JFK, was due to air in the US later this year.
But TV insiders said it was unlikely the series will ever be shown after being branded historically "inaccurate".
Among the most controversial parts of the film were scenes depicting JFK as a "sex fiend" and telling his brother of his need to sleep with women other than his wife.
The series was billed as "an inside look behind the secret doors of the White House," which also "unveils the secrets of the Kennedy family."
It explored the relationship between John F Kennedy (Greg Kinnear), his brother Robert Kennedy (Barry Pepper), and their father Joe (Tom Wilkinson).
Shriver, wife of former California Governor Arnold Schwarzenegger, is alleged to have used her contacts in Hollywood to get the film dropped by the History Channel, which is owned by Disney.
Caroline Kennedy, the daughter of JFK, also allegedly used her connections to persuade History Channel executives to shelve the project.
She is said to have used a forthcoming book about her father that is being published by a Disney subsidiary as leverage with Disney executives.
Even while Holmes and Kinnear filmed the series in Canada last year former Kennedy adviser, Theodore Sorensen, attacked the project calling it "vindictive."
A spokesman for the History Channel said: "While the film is produced and acted with the highest quality, after viewing the final product in its totality, we have concluded this dramatic interpretation is not a fit for the History brand.
"We recognise historical fiction is an important medium for storytelling and commend all the hard work and passion that has gone into the making of the series, but ultimately deem this as the right programming decision for our network."
The series is due to be shown in Canada in March and could still be sold to the British and European stations.
Big deal the man was not a saint! Anyone who has looked at his life and death knows this and i think blocking this is the right thing to do. The powers that be don't want us to worship him and they have very good reasons to slander his name.
