I think, if a family of Nessies are living in the lake - 51 members or so - then they must be creating enough traffic in the water - leaving "shedded skin cells" and defecating and stuff - in the water, that could be collected and analyzed just as a simply, as a spring water sample is analyzed for purity in a laboratory.
The mysterious "crime scene" suddenly becomes a closed Soup Environment, from where the DNA is unable to escape. Hopefully stray "dinosaurian DNA" won't be able to decompose, because it is always being replenished by Nessies merrily paddling around there:
